Abstrak : Salbutamol sulphate is a selective b2 adrenoceptor agonist compounds that undergo a process of first pass metabolism and has a low bioavailability ( 40 % ) in the oral use . One effort to boost the bioavailability of salbutamol sulphate is to formulate the mucoadhesive buccal patches dosage forms . Making the buccal patch is done by using the solvent casting method , using mucoadhesive polymers Na - CMC and PVP K - 30 . This study aims to determine the release of salbutamol sulphate , physical characteristics , and irritation of the mucoadhesive buccal patches with variations in the base levels of Na - CMC and PVP K - 30 . Buccal patch preparations were made with five levels of variation which formula I 100 % : 0 % ; formula II 75 % : 25 % ; formula III 50 % : 50 % ; formula IV 25 % : 75 % ; and the formula V 0 % : 100 % . Patch test results showed the combination of mucoadhesive polymers Na - CMC and PVP K - 30 affect the release of salbutamol sulphate , high concentrations of Na - CMC and the concentration of PVP K - 30 low can boost drug release . Formula II with a concentration ratio of Na - CMC : PVP K - 30 ( 75 % : 25 % ) showed the highest release the number of release 92.19 % . The combination of mucoadhesive polymers Na - CMC and PVP K - 30 affects the physical properties of the buccal patch , the higher the concentration of Na - CMC , the weight , swelling index , the greater thickness . Mucoadhesive buccal patches of salbutamol sulphate produced does not irritate the buccal mucosa with a total production of mucus produced less than 15 % .
