Abstrak : Therapeutic Drug Monitoring (TDM) required to know the level of drug in biological fluid, especially in drug with narrow therapeutic index then can be used to anticipate an under dose and overdose. Digoxin is one of a drug for heart failure who have narrow therapeutic index. The range therapeutic of digoxin 0,8 ? 2,0 ?g/L with half time 48 hours. The fraction of absorption (F) in tablet of digoxin 0,7 with an initial dose 1 ? 1,5 mg/ 70 kg and maintenance dose 0,25 mg/day. In addition blood, can also be used as specimen urine sample in TDM. Factors that determine urine drug faction is using data which is excreted through the urine of at least 10 percent. Digoxin is excreted through the urine about 60-70 percent in the form unchanged drug. The aim of this research is to set levels digoxin in the urine in vitro and knowing parameters validation done already meet the criteria set Food Drug Administration (FDA) in Guidance for Industry: Bioanalytical Method Validation 2001. Preparation of samples using extraction liquid-liquid with diclorometane n-hexan solvent. The analysis method was used HPLC-UV with mobile phase acetonitrile aquabides (35 : 65), stationary phase C18 (5?m.; 4.6 x 150 mm) and UV detector at wavelengths 220 nm. The results standard curve a range between 0,2 to 5 ?g /mL show that this method have linearity r = 0,99995, Limit of detection (LoD) 0,056 ?g /mL, Low Limit of Quantification (LLoQ) 0,085 ?g /mL, and Limit of Quantification (LoQ) 0,17 ?g /mL. The precision of digoxin (CV) 0,222 to 1,789%. The selectivity of digoxin (%diff ) -0,885 to 4,151 %. The accuracy of digoxin (% diff ) -4,690 to 2,364 %. The recovery of digoxin (% recovery) 96,654 to 101,469 %. Digoxin levels in human urine samples can be used to development HPLC-UV which has been validated method.
