| Abstrak | : | Digoxin is a drug with narrow therapeutic index. About 50-70 % digoxin is excreted via urine, thus the concentration can be determined in urine sample. On the development of analytical methods to determined the stability of digoxin to see the influence of temperature and the duration of saving. Analytical method that used High Pressure Liquid Chromatography (HPLC) with ultraviolet detector for digoxin-concentration stability assay in urine sample. The aim of this study was to investigate stability of digoxin stock solution and the digoxin stability in urine sample. The condition of HPLC-UV that use was C18 (5 ?m: 4,6 ? 150 mm) coloumn, mobile phase was aquabidestillata : acetonitril (65:35), and the urine sample was extracted with liquid-liquid extraction technique using dichloromethane : n-hexane (70:30). The result of testing shows that solution digoxin stable at storage for 24 hours (in 25oC) and a week (in 4oC) with te value of % diff [ 7%. A solution of a urine sample containing digoxin the results of the extraction of storage also stable at storage autosampler, short term (in 25oC), freeze-thaw cycle (in -20oC and 25oC), and long term (in -20oC) each reflects the % diff didn?t stray from +15% and -15% Key words : digoxin, urine, liquid-liquid extraction, HPLC-UV, stability |
