| Abstrak | : | Antibiotics resistance is one of serious health problem that lead some researches to find more potent antibacterial agent. One of the sources for new antibacterial agent is traditional medical plants. In central java, the sap of Musa paradisiaca var. sapientum, known as pisang ambon, traditionally is used to prevent infection. Previous research showed that the antibacterial activity of ethanol 96% extract of the midrib of Musa paradisiaca var. sapientum was more active than bark extract against Staphylococcus aureus. Both extract were obtained by maceration using ethanol 96%. In this study, we aimed to evaluate the antibacterial activity against Staphylococcus aureus of the midrib extract of Musa paradisiaca var. sapientum obtained by maceration method using gradient solvents with n-hexane, ethyl acetate, and ethanol 70%, and to determine the minimum inhibition concentration (MIC) and Minimum bactericidal concentration (MBC) of the most active extract. The antibacterial screening was undertaken using agar disc diffusion method, meanwhile the MIC and MBC were determined by dilution test. The result showed that only ethanol extract has antibacterial activity against Staphylococcus aureus with MIC and MBC values of 6.25% (b/v) and 12.5% (b/v), respectively. In addition, phytochemical screening indicated that the ethanol extract contains flavonoid and saponin.
