| Abstrak | : | Rifampicin is a first-line antituberculosis drugs and the drugs that are autoinducer to enzyme cytochrome P-450 monooxygenase 3A4 (CYP3A4). The leaves of gotu kola (Centella asiatica (L) Urban) is empirically has properties such as antituberculosis, this activity shown by compounds asiaticoside. Asiaticoside is an inhibitor of CYP3A4 enzyme. This study aims to determine the effect of oral pretreatment of infusion Centella asiatica leaves against rifampicin on the pharmacokinetics profile in wistar strain male rats. Animal models using mice which consist of two groups, each group consisted of 5 rats and then the rats were fasted for 12 hours and then weighed and treated. Group I (control group) rats were given rifampicin at dose 55 mg/kg BW. Group II (treatment group) rats were given a pretreatment infusion gotu kola leaves 1,89 g/kg BW and rifampicin 55 mg/kg BW. In the hour-0,25; 0,5; 1; 1,5; 2; 3; 4; 5; 6; 8; 10; 12; 21 and 24, blood samples were taken and then set the levels of rifampicin in the blood sample by HPLC. The result obtained made curve rifampicin levels in the blood over time is then determined value rifampicin pharmacokinetic parameters such as Cmax, tmax, Vd, ClT, t1/2, k, AUC0-8, AUMC, MRT, AUC0-24 and AUMC0-24. Pharmacokinetic parameters obtained showed that the administration of Centella asiatica leaf infusion pretreatment can affect the absorption of rifampicin.
