| Abstrak | : | Noni (Morinda citrifolia L.) is one kind of plant that is commonly used empirically as tuberculosis treatment and can inhibit the growth of Mycobacterium tuberculosis strains sensitive or resistant. Noni juice is known to induce drug biotransformation, mainly cytochrome P450 3A4, and rifampicin is a substrate of CYP 3A4. This research aimed to determine the effect of noni juice on the pharmacokinetics profile of rifampicin administered orally to male Wistar rats. Animal model using rats that consists of 2 groups, each group consisted of 5 rats then rats fasted for 12 hours and then weighed and treated. For group I, rats were given a dose of rifampicin 55 mg/kg BW as a control group. Group II, rats were given pretreatment noni juice 4 mL/kg BW for 7 days and consecutively given rifampicin 55 mg/kg BW as the treatment group. On 0,25; 0,5; 1,0; 1,5; 2; 3; 4; 6; 8; 10; 12, 21; and 24 hours blood sample was taken and rifampicin levels in blood samples were analyzed by HPLC. The results obtained made the curve of rifampicin plasma level concentration versus time and then determined pharmacokinetic parameters of rifampicin such as Cmax, Tmax, Vd, CIT, t1/2, k, AUC0-8, AUMC, and MRT. Pharmacokinetic parameters obtained in the two treatment groups were analyzed by independent t-test. The results showed a significant decrease in CLT (p [0.05) by 36,54% (p[0,05), resulting in an significant increase AUC0-8 by 66,07% (p[0,05). Keywords: pharmacokinetics, noni, rifampicin |
