Abstrak : Duwet bark (Syzygium cumini) has been widely studied as an anti-inflammatory and anti-allergy. Duwet bark contains gallic acid, flavonoids, and tannins. All of these three compounds are known to inhibit the release of histamine and proinflammatory cytokine production in mast cells. Inhibition of histamine release can prevent contraction in the treatment of asthma which is mainly caused by allergies. The purposes of this study were to investigate the activity of ethanolic extract of duwet bark as antihistamines for antiasthma alternative medinine, and to determine the mechanism of antihistamine activity of duwet bark ethanolic extract, which were influenced by calcium dependent or independent on guinea pig isolated trachea. The extract of duwet bark was obtained by maceration using 95% ethanol. The method to observe the effect of the extract was isolated organ method with the help of a set of organ bath. Contraction responses were recorded and converted into curve which showed the relationship between the percentage (%) of smooth muscle contraction response vs. logarithm of the agonist concentration. Data were analyzed using ANOVA. The results showed that the ethanolic extract of the duwet bark with concentration of 200 ?g/ml which has pD2 value of 4.37 (p=0.026) decreased the contraction of tracheal smooth muscle significantly, while contraction was decrement at concentration of 50 ?g/ml [pD2 4.59 (p=0.262)] and 100 ?g/ml [pD2 4.52 (p=0.130)] which were not significantly different compared with controls. Logarithmic curve of histamine concentration vs. percent contraction response indicated that there was no reduction in the maximum response on each extract dose given. Reversibility test showed pD2 values at doses of 50 ?g/ml, 100 ?g /ml, and 200 ?g /ml were 4.94 (p=0.999); 4.79 (p=0.730); and 4.75 (p=0.560), respectively. These values did not differ significantly compared to histamine control with pD2 value of 4.96. Based on these result it can be concluded that the ethanolic extract of duwet bark with concentration 200 ?g/ml was H1 antagonist with competitive antagonist type and reversible. Ethanolic extract of duwet bark was calcium independent which was proved by p value on each two minutes measurement at the twelfth to twentieth minutes, which were 0.020; 0.016; 0.018; 0.010; and 0.010.
