| Abstrak | : | Vitex trifolia Linn. or legundi leaves is one of the traditional medicine to treat tuberculosis. Diterpenoid compounds of legundi known to inhibit the growth of Mycobacterium tuberculosis. This research aimed to determine the effect of legundi leaves infuse on the pharmacokinetics profile of rifampicin administered orally to male Wistar rats. In this study, animal trials were divided into two groups, each group consists of 5 rats. The group I were given a single dose of rifampicin 55 mg/kgBB as a control, while the group II were given single doses of legundi leaves 2100 mg/kgBB orally once daily for seven days. On eight day was given concurrently with rifampicin dose 55 mg.kgBB. Blood samples were taken at 0,25; 0,5: 1; 1; 2; 3; 4; 16; 8; 10; 12; 21 dan 24 hours. Determination of rifampicin levels in plasma were analyzed by High Performance Liquid Chromatograpy (HPLC). The result obtained created rifampicin curve in blood toward time, then determined pharmacokinetic parameters such as Vd, Cl, t ?, k, AUC, AUMC, MRT, tmaks, and Cpmaks. The result showed pretreatment infuse of legundi leaves may decrease the elimination of rifampicin indicated by decrease ClT by 31,20% (p[0,05). Decrease ClT value cause increase AUC 39,87% and Cpmaks 51,66% (p[0,05). Keywords : pharmacokinetics, legundi leaves, rifampicin, HPLC |
