Abstrak : Tobacco (Nicotiana tabacum) is a crop plantation widely used as the raw material of cigarettes manufacturing. Based on previous research, the extract of tobacco leaves has antibacterial activity, therefore tobacco leaves extract might be applied in a pharmaceutical dossage form as antiseptic hand sanitizer gel. This studies used carbopol940 and Triethanolamine (TEA) that often were used in the form of gel. The aims of these studies were to determine the optimum formulation of carbopol940 and TEA in various concentration and to evaluated antibacterial activity antiseptic hand sanitizer gel from the extract of tobacco leaves with formula optimum. The extraction of tobacco leaves was using maceration with ethyl acetate as solvent. The optimum formulation for carbopol940 and TEA was obtained by using Simplex Lattice Design (SLD). Physical stability test of the gel includes organoleptic, adhesion, dispersion, viscosity, and pH. Antibacterial activity of tobacco leaves extract was tested by disk diffusion method (Kirby-Bauer), while the antibacterial activity of hand sanitizer gel was examined by hole-plate diffusion method at weeks 0 and 4. The tobacco leaves extract in hand sanitizer gel has MIC for the Eschericia coli and Staphylococcus aureus at concentration of 0.125 mg/?l. The result of the optimum formula from SLD was the formulation with ratio 0.4g : 0.9 g of carbopol 940 and TEA. The result of stability test during four weeks from the optimum formula gel of tobacco leaves extract showed decreasing in viscosity. The zone of inhibition was higher at week 4, because the tobacco leaves extract required time to be able to dissolve in the form of gel so that the effectivity of the antibacterial became higher.
